LB Medium is a rich medium for the growth of E. coli and certified for plasmid and other bacterial cultures in liquid medium. The two main components of LB media are Tryptone and Yeast Extract. Tryptone is used in a concentration of 10 gram / liter and Yeast Extract in a concentration of 5 gram / liter. LB medium (Miller) is the high salt LB medium type with 10 gram /liter. Media Preparation: Reagents are tested and chosen for their ability to promote optimum growth of E. coli when combined in LB Medium formulations. Chosen reagents are combined in the accurate proportions according to the LB formulation and thoroughly blended for a uniform distribution. A sample of the blended formulation was used to prepare the liquid. The combinations were mixed to dissolve components and then autoclaved at 121°C for 15 minutes.
